ABSTRACT

OBJECTIVE:To establi sh the quality standard of Yao medicine Pileostegia tomentella,and to provide reference for its quality control. METHODS :Totally 10 batches of P. tomentella were collected from Guangxi area. The properties of P. tomentella were observed and microscopic identification was conducted for the transverse section of its stems ,roots and powder. TLC method was established by using 7-hydroxycoumarin as reference. The contents of water ,total ash ,acid-insoluble ash and alcohol-soluble extract in P. tomentella were determined. The contents of skimmin ,7-hydroxycoumarin and 7-hydroxy-8- methoxycoumarin in P. tomentella were determined by HPLC. RESULTS :The root and stem surface of P. tomentella possessed obvious wrinkles ,hard texture ,slight smell and bitter taste ,and there were secretory cells and needle crystal cells in the cross section. The medicinal powder contained calcium oxalate needle crystal ,calcium oxalate square crystal ,sclerotic cell and starch granules. The results of TLC showed that the spots of the same color were found in the corresponding positions of chromatograms of test samples and 7-hydroxycoumarin control. In the 10 batch of P. tomentella ,the contents of water were 9.1%-11.8%;those of total ash were 3.6%-6.7%;those of acid-insoluble ash were 0.10%-0.44%;those of alcohol-soluble extract were 8.9%-14.0%.The linear range of skimmin ,7-hydroxycoumarin and 7-hydroxy-8-methoxycoumarin were 6.088-182.640,1.568-47.040 and 1.096- 32.880 μg/mL(all r not less than 0.999 7). The average recoveries were 99.47%,101.07% and 99.89%(RSD were all less than 2%). RSDs of precision ,stability(24 h),reproducibility and durability tests were all lower than 2% . The contents of 3 components such as skimmin were 1.337-9.534,0.348-2.236, 0.083-1.088 mg/g,respectively. CONCLUSIONS :It is preli- 201705) minarily proposed that the content of water in P. tomentella is not more than 12.0% ;that of total ash is not more than 7.0%;that of alcohol-soluble extract is not less than 8.0%; E-mail:luguoshou@foxmail.com that of ski mmin is not less than 1.30 mg/g;that of 7-hydroxy- coumarin is not less than 0.30 mg/g;that of 7-hydroxy-8-methoxycoumarin is not less than 0.06 mg/g.

ABSTRACT

Lung cancer is the leading cause of cancer death and the most common malignant tumor, the long-term survival of which has stagnated in the past several decades. Pileostegia tomentella Hand. Mazz is a traditional Chinese medicine called "Zhongliuteng" (ZLT) in the pharmacopeia, which has been proved to possess a potent anti-tumor effect on various cancers. In this study, the effects of ZLT N-butanol extraction (ZLTN) and ZLT ethyl acetate extraction (ZLTE) on the viability of non-small cell lung cancer cell (NSCLC) lines H1299 and A549 were evaluated. Here, we firstly reported that ZLTE significantly inhibited H1299 cells growth without affecting the release of lactate dehydrogenase (LDH). In addition, ZLTE induced caspase-dependent apoptosis in a concentration-dependent manner and increased the expression cleaved-PARP and decreased pro-caspase-3, pro-caspase-7, pro-caspase-8, and pro-caspase-9. Moreover, ZLTE increased the level of cellular reactive oxygen species (ROS) in H1299 cells to lead to apoptosis, which was reversed by N-acetyl-cysteine (NAC). Taken together, our results revealed that ZLTE induced caspase-dependent apoptosis via ROS generation, suggesting that ZLTE is a promising herbal medicine for the treatment of NSCLC.

ABSTRACT

OBJECTIVE: To study the chemical constituents of Yao medicine, Zhongliuteng, the stems of Pileostegia tomentella. METHODS: The chemical constituents were isolated and purified by silica gel chromatography repeatedly, and their structures were identified by spectral analysis and chemical METHODS. RESULTS: Thirteen compounds were isolated from the stems of P. tomentella and the structures were identified as 1-O-(β-D-glucosyl)-2-[2-methoxy-4-(ω-hydroxypropyl) phenoxy]propan-3-ol(1), (+)-lyoniresinol-3a-O-β-D-glucopyranoside (2), syringin (3), coniferin (4), dihydroconiferin (5), but-3-enyl-β-D-glucoside (6), 4-(2, 3-dihydroxypropyl)-2, 6-dimethoxy phenyl β-D-glucopyranoside (7), nikoenoside (8), protocatechuic acid ethyl ester (9), 8-methoxy coumarin-7-O-β-D-glucopyranoside (10), 6-O-R-L-rhamnopyranosyl-β-D-glucopyranoside methyl salicylate (11), nicotinamide (12), and 3, 5-di-O-caffeoyl quinicacid methyl ester (13). CONCLUSION: All compounds were obtained from the genus for the first time.

ABSTRACT

Objective: To study the chemical constituents of Yao medicine Zhongliuteng, the canes of Pileostegia tomentella. Methods: The chemical constituents were isolated and purified by silica gel chromatography repeatedly from the canes of P. tomentella and their structures were identified by spectral analysis and chemical methods. Results: Sixteen compounds were isolated from the canes of P. tomentella and their structures were identified as loganic acid (1), 4-O-α-L-arabinofuranosyl-(1→6)-β-D-glucopyranoside (4S)-4-hydroxyl-β-ionone (2), resorcinol (3), daphnin (4), skimmin (5), umbelliferone (6), vogeloside (7), diethyl phthalate (8), secologanin (9), secologanin dimethyl acetal (10), sweroside (11), foliasalacioside B (12), benzyl-O-β-D-apiofuranosyl-(1″→6')-β-D-glucopyranoside (13), maltose (14), glucose (15), and daucosterol (16). Conclusion: All compounds are obtained from the plant for the first time, and all compounds are also obtained from the plants of Pileostegia Hook. f. et Thoms. for the first time except compounds 5, 6 and 16.

ABSTRACT

OBJECTIVE: To study the antitumor effect of Pileostegia tomentella 95% alcohol extract (PTAE) on H22 tumor-bearing mice and its possible mechanisms. METHODS: Sixty mice were chosen and mouse models bearing H22 solid tumor were established in fifty mice, and the others were as normal control. H22 tumor-bearing mice were randomly divided into five groups:model control group, fluorouracil group(20 mg·kg-1), PTAE high, middle and low-dose group(180, 90, 45 g·kg-1 of crude drug, respectively). The mice in treatment groups were intragastric administration respectively, meanwhile, the mice in normal control and model groups were treated with the same volume of distilled water, once a day for ten days. The blood was collected from eyeball in all mice, and the serum were separated and detected by ELISA for IL-2 and TNF-α. Then the mice were put to death. Their tumors, thymuses and spleens were separated and weighted, and the tumor inhibitory rates, thymus and spleen indexes were calculated. The pathological change of tumor tissue was observed. RESULTS: Compared with model control group, the tumor weights of PTAE high and middle-dose groups were significantly decreased (P<0.05, P<0.01), the tumor inhibitory rates were 37.44% and 38.46% respectively. The spleen index of PTAE middle-dose group was increased significantly (P<0.01). The level of IL-2 in serum of tumor-bearing mice in the PTAE high-dose group was increased significantly(P<0.05), and the level of TNF-α in serum (P<0.01) could be increased significantly in the PATE high, middle and low-dose groups. CONCLUSION: Pileostegia tomentella 95% alcohol extract has antitumor activity, its mechanism may be developed by immuno-regulation.
